Subculture	Nutritional gravy medium (English name: NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (not included in liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mgMnSO4 · H2O to culture Bacillus is beneficial to produce spores.
Subculturing	Before use, the reference product is placed at room temperature for 30min, fully melted, shaken and mixed, used after nucleic acid extraction, nucleic acid extraction or purification reagents need to be provided by oneself.

Application	This reference is a heat-inactivated Escherichia coli phage PhiX174. The host bacteria of Escherichia coli are cultured, and phage PhiX174 are inoculated to collect phages. The collected phage titer and lysis efficiency were determined, and double-layer agar was used for counting. The heat-inactivated reference has no infection and pathogenic ability, but retains the complete genetic material of the virus particles. Mainly used for quality control substances and positive controls in related nucleic acid detection processes.
