| Subculture | Nutritional gravy medium (English name: NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (not included in liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mgMnSO4 · H2O to culture Bacillus is beneficial to produce spores. |
| Subculturing | Samples require a total of 200mL of sterile water or sterile saline to dissolve. Before use, the quality control sample is placed at room temperature for 30min, then opened in the biosafety cabinet, 3~4mL sterile water is immediately added, dissolved and sucked out into a sterile bottle, and the remaining sterile water is repeatedly washed into the inner wall of the Xilin bottle. All transferred to the above sterile bottle is negative quality control sample, totaling 200mL, with a concentration of 300 ~ 3000MPN/L. |
| Growth conditions | Culture temperature 37 ℃; Culture time 24-48 hours; Aerobic gas environment |
| Storage conditions | -20 ℃, kept away from light |
| Safety level | 1 |
| Application | It is suitable for the determination of fecal coliforms in water quality, quality control and personnel assessment (multi-tube fermentation method). Reference Standard "HJ 347.2-2018 Water Quality Determination of Fecal Coliforms Multi-tube Fermentation Method" |
