Subculture Tryptone soybean medium (English name: TSA/TSB): tryptone 17.0g, soybean peptone 3.0g, sodium chloride 5.0g, dipotassium hydrogen phosphate 2.5g, glucose 2.5g, agar 20.0g (without liquid medium), distilled water 1.0L,pH 7.3±0.2. Sterilization at 121 ℃ for 15min.
Subculturing 1. Before disinfection, the bacteria tablets shall be placed on the disinfection evaluation site according to the distribution requirements; 2. After disinfection reaches the action time, transfer the bacteria tablets into a test tube equipped with 5.0mL of corresponding neutralizer with sterile tweezers, vibrate 80 times in the palm of the hand or mix evenly with a mixer, and neutralize for 10min. 3. Set up positive control group. During low temperature on-site disinfection, the positive control group and the test group were put into the corresponding low temperature environment together, and after reaching the same low temperature, they were put into the diluent. 4. Oscillate the sampling tube on the mixer for 20s or beat it 80 times on the palm of the hand, absorb 1.0mL of the sample to be tested and inoculate it into a sterile plate, inoculate 2 plates in parallel for each sample, and inoculate and culture with corresponding culture medium according to the standard requirements. 5. Count the number of colonies and calculate the killing rate.
