Subculture Nutritional gravy medium (English name: NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (not included in liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mgMnSO4 · H2O to culture Bacillus is beneficial to produce spores.
Subculturing 3~5 single colonies were selected from the plate and mixed with 5mL sterile PBS to prepare bacterial suspension. Take 100 μL of bacterial suspension and inoculate each trace of vials and test tubes (solid medium directly pick single colony inoculation). The inoculated vials and test tubes were then covered with rubber stoppers, cultured with semi-plug, erected in the groove of the inner support, or placed in a suitable bottle holder, cultured in an incubator at 35-37 ℃ for 24-48h, and the results were interpreted with reference to "GB4789.30-2016 Food Microbiological Examination Listeria monocytogenes Test.
