| Subculture | Nutritional gravy medium (English name: NA/NB): beef paste 3.0g, peptone 10.0g,NaCl 5.0g, agar 20.0g (not included in liquid medium), distilled water 1.0L,pH 7.0. Sterilization at 121 ℃ for 15min. [Note] Adding 5 mgMnSO4 · H2O to culture Bacillus is beneficial to produce spores. |
| Subculturing | Samples require a total of 500mL of sterile water or sterile saline to dissolve. Before use, the quality control sample is placed at room temperature for 30min, then opened in the biosafety cabinet, 3~4mL of sterile water is immediately added, dissolved and sucked out into a sterile bottle, the remaining sterile water is repeatedly washed into the inner wall of the Xilin bottle, and all of them are transferred to the above sterile bottle, which is a negative quality control sample with a total concentration of 500mL and a concentration of 40 ~ 600CFU/L. |
| Growth conditions | Culture temperature 37 ℃; Culture time 24-48 hours; Aerobic gas environment |
| Storage conditions | -20 ℃, kept away from light |
| Safety level | 1 |
| Application | Suitable for water quality fecal coliform determination quality control and personnel assessment (filter membrane method). Reference Standard "HJ 347.1-2018 Water Quality Determination of Fecal Coliform Bacteria Filter Method" |
| Sharing mode | Public welfare sharing |
